Action of plant proteinase inhibitors on enzymes of physiopathological importance Anais da ABC (AABC)

Obtained from leguminous seeds, various plant proteins inhibit animal proteinases, including human, and can be considered for the development of compounds with biological activity. Inhibitors from the Bowman-Birk and plant Kunitz-type family have been characterized by proteinase specificity, primary structure and reactive site. Our group mostly studies the genus Bauhinia, mainly the species bauhinioides, rufa, ungulata and variegata. In some species, more than one inhibitor was characterized, exhibiting different properties. Although proteins from this group share high structural similarity, they present differences in proteinase inhibition, explored in studies using diverse biological models.

Adaptation of digestive enzymes to dietary protein, carbohydrate and fibre levels and influence of protein and carbohydrate quality in Penaeus vannamei larvae (Crustacea, Decapoda) ArchiMer

Activity of digestive enzymes (trypsin, chymotrypsin and amylase) was studied in relation with food in Penaeus vannamei larvae first stages. Microparticles containing casein involved the decrease of soluble protein content and enzyme activities. Specific trypsin activity but not chymotrypsin was correlated (p<1%) to casein level between 10 and 60% in microparticles. Cellulose fibres in food seemed to have little effect on digestive enzymes. Starch, between 1 and 20% in microparticles, had no influence on specific amylase activity. The effect of different feeding conditions were also tested on growth. Characterization and expression analysis of chymotrypsin after bacterial challenge in the mud crab, Scylla paramamosain Genet. Mol. Biol.

Chymotrypsin is one of the serine proteases families that have various biological functions. A chymotrypsin gene was isolated from hepatopancreas of the mud crab, Scylla paramamosain (designated SpCHY) in this study. The full-length cDNA of SpCHY contained 942 nucleotides with a polyadenylation sequence and encoded a peptide of 270 amino acids with a signal peptide of 17 amino acids. The SpCHY gene contains seven exons, six introns, a TATA box and several transcription factor binding sites that were found in 5'-promoter region which is 1221 bp in length-time quantitative PCR analysis indicated that the expression level of SpCHY mRNA in hepatopancreas was significantly higher than that in other tissues. Chymotrypsin- and trypsin-like activities secreted by the multidrug-resistant yeasts forming the Candida haemulonii complex Anais da ABC (AABC)

Abstract: Candida haemulonii complex (C. haemulonii, C. haemulonii var. vulnera and C. duobushaemulonii) consists of emergent multidrug-resistant pathogens that cause bloodstream and deep-seated infections. However, little is known about their virulence factors. Herein, we evaluated the presence of extracellular serine peptidases in this fungal complex. Serine peptidase activity was measured by spectrophotometry using chromogenic peptide substrates to the S1 family. Chymotrypsin-, trypsin- and elastase-like activities were detected in all fungal isolates. Since higher chymotrypsin- and trypsin-like activities were observed from the cleavage of N-succinyl-Ala-Ala-Pro-Phe-pNa and N-benzoyl-Phe-Val-Arg-pNa, respectively, these substrates were selected for...

Enzyme cleavable and biotinylated photoaffinity ligand with diazirine OAK

The efficient synthesis of an enzyme cleavable biotinylated diazirinyl photoaffinity ligand is described to allow the effective manipulation of the photolabeled biocomponents. The compound contains a glutamic acid gamma-methyl ester, which is a precursor of the substrate for V8 protease, between the diazirinyl photophor and biotin moiety. After alkaline hydrolysis of the ester, the compound can be proteolyzed at the Glu moiety by V8 protease. The photophore was introduced to L-Phe p-nitroanilide and the ligand was applied to photolabel of chymotrypsin to manipulate the application of the concept.

Role of Chicken Pancreatic Trypsin, Chymotrypsin and Elastase in the Excystation Process of Eimeria tenella Oocysts and Sporocysts OAK

The role of pancreatic proteolytic enzymes in the excystation process of Eimeria tenella oocysts and sporocysts was studied in vitro. Intact sporulated oocysts were preincubated in phosphate buffer, NaCl 0.9% (PBS) added with 0.5% chicken bile extract in a 5% C02 atmosphere for 30 minutes prior to exposure to either 0.25% (w/v) chicken trypsin, chymotrypsin, pancreatic elastase, or a 1% (w/v) crude extract of unsporulated and sporulated oocysts of E. tenella (Expt.1). Variations des activites digestives en fonction des facteurs du milieu chez les crustaces ArchiMer

Proteolytic enzymes of hepatopancreas of six species of peneids (Penaeus kerathurus, P. japonicus, P. monodon, P. stylirostris, P. merguiensis and P. vannamei ) have been studied. Some of their physiological variations have been characterised. Increase in acclimation temperature of crustaceans enhances increase of specific activities of proteases. Variations of salinity does not modify specific activities, nor quantities of trypsin, nor electrophoretic patterns of these different proteases.
